$\begingroup$ I wish I could vote more. I think RegionMember is by far the fastest based on my test. Who stands with me ! I wish you could add a "Canada test" as Daniel Lichtblau has done. That will better show the power of RegionMember when dealing with complex polygons and lots of points. With a pregenerated RegionFunction at first, RegionMember beats other method badly. $\endgroup$
